In Angry Birds Rio, Red, Chuck and Blues are kidnapped and taken to Rio by smugglers under the orders of an evil cockatoo parrot named Nigel. As in the original Angry Birds, players throw birds with slingshots to try to hit targets on or inside structures. Instead of pigs stealing eggs, they must rescue exotic birds in the cage or, depending on the level played, defeat Nigels marmosets. Many characters from Rio stand out. Blu and Jewel stand out as game-specific bird species along with all existing birds in the series. The game also features players first-ever boss battles in the series to defeat Nigel, the main antagonist in the movie, and Mauro, the leader of the marmosets.

Angry Birds Rio has the same gameplay as the first Angry Birds game. Players take control of a group of birds that can be launched anywhere using a slingshot. All levels have a number of enemies, which can be caged birds or marmosets that the player must defeat using their characters. These enemies normally appear sheltered inside structures made of wood, glass and/or stone, along with some additional items such as chains and TNT. If the players cannot kill all the enemies in the level, they are given a chance to try again, if they succeed, they win and move on to the next level. One, two or three stars are awarded depending on the score obtained at each level.

Like Angry Birds, Angry Birds Rio consists of several parts. Each level can be unlocked after completing the previous one. After the release of Rio 2, the episodes were divided into two different categories: Rio and Rio 2. Episodes in Rio have Angry Birds Chrome-independent images for birds and have the same mechanics as Angry Birds. On the other hand, episodes in the Rio 2 category contain images that preserve the appearance of the birds in Angry Birds Toons. Additionally, there are optional hints at the levels in these chapters. Gameplay does not change in all levels. There is also a tutorial section that shows players the most basic mechanics of the game.

The levels in Angry Birds Rio consist of various levels; Episodes in Rio have thirty levels, while episodes in Rio 2 have twenty levels. Additionally, all levels except Playground and Hidden Harbor have six extra optional bonus levels that can be unlocked by completing certain missions or spending coins.

Angry Birds Rio, like other Angry Birds games, has optional power-ups that players can use to gain a temporary advantage, allowing them to pass the level more easily. These buffs are limited. There are in-game currency (coins) available that allow players to buy more power-ups and unlock bonus levels. Coins can be obtained by collecting stars or watching advertisements.
